| 目的研究30nm胶体金对小鼠成骨细胞系MC3T3-E1增殖、分化及成骨相关基因RUNX2表达的影响。方法体外培养小鼠成骨细胞MC3T3-E1，在仪-MEM培养液中加入不同体积的胶体金处理细胞，利用M1Tr法检测胶体金对成骨细胞增殖活性的影响，碱性磷酸酶活性测定及RT—PCR法分别检测其对成骨细胞分化及成骨相关基因RUNX2表达的影响。结果浓度为10-10、10-11、10-12mg/L的胶体金均促进成骨细胞的增殖，且ALP相对活性及RUNX2基因表达也均高于阳性对照组。结论30nm胶体金能促进小鼠前成骨细胞系MC3T3-E1增殖、分化及成骨相关基因RUNX2的表达，进而促进成骨。 |

| Objective To investigate the effect of gold colloidals （Au NPs） on proliferation, differentiation and expression of runt-related transcription factor 2 （Runx2） in MC3T3-E1 cells. Methods Mouse calvarial preosteoblasts （MC3T3-E1） were cultured on alpha minimum essential medium （α-MEM） and treated with different concentrations of gold colloidals. The cell proliferation was evaluated by MTT method, cell differentiation was assessed by measuring alkaline phosphatase （ALP） activity and the expression of RUNX2 mRNA was detected by reverse transcriptase polymerase chain reaction （RT-PCR）. Results Gold colloidals at concentrations of 10-10、10-11、10-12 mg/L promoted cell proliferation and differentiation and enhanced the expression of RUNX2 mRNA in MC3T3-E1 cells. Conclusion Results indicate that gold colloidals can enhance cell proliferation and osteogenic differentiation and Runx2 expression in MC3T3-E1, indicating that it may have potential to promote bone regeneration. |
