纳米氟磷灰石/聚醚醚酮种植体成骨效能研究

摘要:
目的探索纳米氟磷灰石/聚醚醚酮(nFA/PEEK)复合材料的生物相容性及成骨效能。方法将研发喷砂处理的nFA/PEEK和聚醚醚酮(PEEK)种植体植入Beagle犬双侧胫骨,于植入第8周和12周末获取骨组织标本并且行大体观察和X线摄片检查。带种植体不脱钙骨磨片行苦味酸品红染色、测量骨结合率(bone implantcontacts,BIC);四环素荧光标记测定骨矿化沉积率(mineral apposition ratio,MAR)了解种植体骨界面改建过程。结果nFA/PEEK可X线摄片显影,其种植体周围新骨形成和成熟比PEEK更快,nFA/PEEK种植体在8周末和12周末的MAR和BIC均高于对应的PEEK(P<0.05)。12周末nFA/PEEK种植体组的BIC均高于8周末nFA/PEEK组(P<0.05),而MAR却降低(P=0.019)。nFA/PEEK种植体组12周末的BIC(82.01±3.61)%为组内最高,而8周末MAR是组内最高,为(1.49±0.08)μm/d。结论nFA/PEEK种植体能与骨形成骨性结合,生物相容性和成骨效能良好,具有潜在临床应用价值。

Osteogenesis initiated by nano-fluorapatite polyetheretherketone implants

Abstract:
Objective To investigate the effect of nano-fluorapatite polyetheretherketone (nFA/ PEEK) composites on osteogenesis in dental implants. Methods Sandblasted nFA/PEEK and PEEK implants were performed in both sides of tibias of Beagles dog. Samples of implants and bone tissues were collected in 8th and 12th weeks after implantation, which were examined by X-ray and gross observation. Implants and undecalcified bone tissues were cut and further stained by Van-Gieson method and the bone implant contacts (BIC) were evaluated. The new bone tissues labeled by tetracycline were observed under fluorescent microscopes and the mineralization apposition ratio (MAR) was calculated. Results X-ray revealed that nFA/PEEK implants were observed to embedded in new bones, which was more intensive than that in PEEK's. MAR and BIC in nFA/PEEK at 8th and 12th week were higher than those in PEEK. BIC in nFA/PEEK at 12th week was significantly higher (P<0.05), whereas MAR was smaller than that at 8th week (P=0.019). BIC (82.01±3.61 )% in nFA/PEEK implants at 12th weeks was the highest, while MAR at 8th week (1.49±0.08μm/d) was the highest. Conclusion The study demonstrates that nFA/PEEK implants are osseointegrated with bone beds, indicating its good biocompatibility and potentially clinical applicability.
