腺苷酸环化酶关联蛋白1与肺癌转移及预后的相关性研究

摘要:
目的探讨腺苷酸环化酶关联蛋白1(cyclase-associated protein1,CAP1)在肺癌转移时的表达及与肺癌转移和预后的关系。方法Western印迹法、免疫组织化学法测定肺癌组织与肺癌细胞株在蛋白水平的CAPl表达。并同肺癌的转移和预后进行相关性研究。结果单克隆抗体的Western印迹法检测发现CAPl在肺癌组织有高表达,而正常组织表达较少,发生转移的肺癌组织表达更高。对82例患者进行免疫组织化学分析结果表明,CAP1的阳性率与肺癌转移相关(尤其是淋巴结转移的程度),且CAPl高表达构成肺癌发生转移的独立危险因素。Kaplan—Meier生存分析提示CAPl的高表达预示着不良的预后。结论肺癌CAPl的表达比正常组织高,高表达的CAPl与肺癌转移相关。

Abstract:
Objective To investigate the relationship of adenylate cyclase-associated protein 1 (CAP1) with metastasis and prognosis of lung cancer. Methods The expression of CAP1 in lung cancer tissues was assessed by Western-blotting and immunohistochemistry, respectively. The relationship of CAP1 expression with metastasis and prognosis of lung cancer was analyzed. Results Expressions of CAP1 were high in lung cancer tissues with or without metastasis and low in the noncancerous lung tissues by Western-blotting analysis. And the intensity of the band was stronger in the lung cancer of metastasis. The expression of CAP1 protein in 82 cases of lung cancer was detected by immunohistochemical analyses. The ratio of CAP1 -positive tumor cells in clinical specimens was correlated with the presence of metastasis (especially lymph node metastasis) and overexpression of CAP1 was an independent risk factor for metastasis of lung cancer. Kaplan-Meier survival analysis demonstrated that high level of CAP1 expression in lung cancer was associated with poor prognosis. Conclusion CAP1 is up-regulated in lung cancer and overexpression of CAP1 is correlated with metastasis and poorer prognosis of lung cancer.
