Alternation of HSP60 and trypsin expression in pancreatic tissues in experimental acute pancreatitis

Abstract:
Objective To observe dynamically the alternation of heat shock protein 60(HSP60) and trypsin expressions in the pancreatic tissues with different severities of inflammation in vivo and in vitro,and to explore their potential role in the pathogenesis of acute pancreatitis(AP).Methods In vivo study,the mild acute pancreatitis(MAP) in mice was induced by cerulein intraperitoneal injections six times hourly at a dose of 50μg·kg-1,while normal control animals received injections with an equal volume of normal saline(NS).The animals were killed 3 hrs after the last injection,and the pancreatic tissues were taken for further examination.The severe acute pancreatitis(SAP) in rats was induced by a retrograde injection of 3%sodium deoxycholate into the bilio-pancreatic duct at a dose of 1 ml·kg-1.The control rats received a sham operation without the retrograde injection.The animals were killed respectively after 1 hr,5 hrs and 10 hrs of the SAP modeling and the pancreatic tissues were taken for further examination.In vitro study,rat pancreas were isolated and the pancreatic tissue snips were prepared,cultured, and stimulated with different concentrations of lipopolysaccharide(LPS,10,20μg·ml-1) and cerulein (10-11,10-5 mol·L-1),while the normal control group got NS.The tissue samples were taken 4 hrs after stimulation.All the harvested pancreatic tissues were examined for the pathological changes with HE,and for the alternation of HSP60 and trypsin expressions by immunohistology with double immunofluorescence. Results The pancreatic tissues of MAP in mice showed a mild congestion and edema,and the expressions of HSP60 and trypsin in the pancreatic tissues were significantly higher(P<0.01).The pancreatic tissues of SAP rats showed obvious hemorrhagic and necrotic changes.The expression of HSP60 in the tissues increased 1 hr after the modeling of SAP and maintained at a high level at the time points of 5 hrs,10 hrs after the SAP modeling,and the differences were significant compared with those in the normal control group(P<0.05).While the expression of trypsin in the tissues,compared with that in the control group (P<0.05 ),increased unsignificantly 1 hr after the SAP modeling(P>0.05 ),and then elevated significantly with the extension of modeling time.The expressions of HSP60 and trypsin in the isolated rat pancreatic tissue,compared with those in the normal control group,decreased significantly under the stimulation of LPS or cerulean(P<0.05).Conclusions In the experimental conditions,whether in mild or severe AP,the expression of trypsin in the pancreatic tissues was elevated and the same to the expression of HSP60;however,the increase ranges of HSP60 and trypsin expression in different degrees of severity of pancreatic injury are of different trend.This indicates that the imbalance of HSP60 and trypsin expression may be involved in AP development.The expressions of HSP60 and trypsin may be reduced when stimulating directly the isolated pancreas fragments with different concentrations of LPS and cerulein.
