Abstract:
Objective To evaluate the role of interferon-γin acute pancreatitis in rats.Methods The models of acute pancreatitis were randomly divided into nine groups:acute pancreatitis 6 hour group, 12 hour group,24 hour group,48 hour group,IFN-γ6 hour group,12 hour group,24 hour group, 48 hour group,and control group.Serum,mesenteric lymph nodes(MLN),and pancreatic tissue were harvested.Serum amylase,endotoxin,TNF-α,IL-18,and IL-27 were detected.Pathologic changes of pancreatic tissue were observed.The protein of TNF-α,NF-κB,and IL-18 of macrophage in pancreatic tissue were observed with immunofluorescence dual staining.Results Serum TNF-α, IL-18,and IL-27 all increased in AP groups.IL-18 increased more significantly in INF groups.TNF-αand EL-27 in INF groups slightly decreased compared with those in AP groups,but still higher than those in the control group.The level of serum amylase increased both in AP groups and in INF groups, but increased more significantly in INF groups.Edema and inflammation of pancreatic tissue were observed both in AP groups and in INF groups.Results from the immunofluorescence double staining of macrophages in pancreatic tissue showed that TNF-α,NF-κB,and IL-18 expressed in AP groups and in INF groups,but not in the control group.The interstitial edema,congestion and inflammation of pancreatic tissue were more obvious in INF groups.NF-κB and IL-18 staining were more significant in INF groups.Conclusions IFN-γhas effects on the level of cytokines in acute pancreatitis and can make pancreatitis more severe.Control of IFN-γmay relieve pancreatitis to a certain extent.
