目的探讨血必净注射液对脓毒血症大鼠炎症反应的影响。方法 将100只雄性Wistar大鼠,随机分为正常对照组、假手术组、盲肠结扎穿孔术(cecum ligation and puncture, CLP)组、血必净组,每组25只大鼠,每组分为5个亚组。CLP组及血必净组大鼠均使用CLP建立脓毒血症大鼠模型。血必净组大鼠分别于术后2、12、24、36、48、60h经阴茎背静脉注射血必净注射液(天津红日药业)4ml/kg,其余各组大鼠经阴茎背静脉注射生理盐水4ml/kg。术后6、12、24、48、72h,分别收集各组实验大鼠的双肺。取大鼠左全肺,测定肺湿干重比(W/D);取大鼠右肺,酶联免疫吸附测定(ELISA)法检测各组大鼠肺组织匀浆上清液中IL-17及IL-6浓度。结果 脓毒症组大鼠肺湿干重比、IL-17浓度在术后6h开始出现上升,至术后12h达到高峰,在术后12~72h维持在较高水平。肺湿干重比、IL-17浓度上升均有统计学意义(P<0.05) ,血必净组大鼠IL-17浓度在各个时间节点均低于脓毒症组,差异有统计学意义(P<0.05)。结论 血必净注射液可降低脓毒血症大鼠肺脏组织IL-17浓度,对脓毒血症具有一定的治疗作用。 |

Effect of TCM Xuebijing injection on inflammatory response in rats with sepsis |

Abstract: |
Objective To investigate the effect of TCM Xuebijing injection on the inflammaroty response in rats with sepsis. Methods One hundred male Wistar rats were randomly divided into control group, sham operation group, CLP group, and Xeubijing group with 25 rats in each group. Sepsis was induced with cecum ligation and puncture (CLP) in rats of CLP and Xeubijing groups. Xeubijing (Tianjin Chase Sun Pharmaceutical Co., Ltd.) was injected through venae dorsales penis with a dosage of 4ml/kg at 2,12,24,36,48,60h after surgery in Xeubijing group, while normal saline was injected in rats of other groups. The lungs were harvested at 6,12,24,48,72h after the final injection. The wet to dry weight ratio (W/D) of the lung was measured. The contents of IL-17 in lung tissue homogenate supernatant were measured by enzyme-linked immunosorbent assay (ELISA). Results The W/D of lung and the concentrations of IL-17 in lung tissue supernatants of CLP group were rising 6h after operation, peaked at 12h and maintained at a high level during 12 to 72h (P<0.05). The IL-17 concentrations in Xuebijing group were significantly lower than those in CLP group at all time points (P<0.05). Conclusion TCM Xuebijing injection can reduce IL-17 contents in lung tissue and alliviate the inflammatoty response in rats with sepsis. |
