Abstract:
Objective To investigate the expression of microRNA-182(miR-182) in triple-negative breast cancer(TNBC) and its clinicopathological significance. Methods Thirty patients with triple-negative breast cancer underwent mastectomy, and the surgical specimens were collected. Quantitative Real-Time polymerase chain reaction(RT-PCR) was used to detect the expression of miR-182 in cancer tissues and pericancerous tissue. And the relationship between miR-182 expression in TNBC cancer tissues and clinicopathological characteristics, including tumor size, grade, lymph node metastasis, distant metastases, TNM staging, disease-free survival, distant metastasis-free survival, overall survival were analyzed. ResultsRT-PCR results indicated that the expression of miR-182 in cancer tissue was 8.6 times higher as that in pericancerous tissue (P<0.001). In cases of lymph node positive and TNM stage Ⅲ, the expression of miR-182 was higher than those of lymph node negative(Z=-3.454,P=0.001) and TNM stage(Ⅰ+Ⅱ)(Z=-3.946,P=0.000).Cox proportional hazards risk analysis indicated that the high expression of miR-182(HR=0.107,5% CI:0.018-0.643,P=0.015), positive lymph nodes(HR=0.189,5% CI:0.020-1.757,P=0.016), lymphovascular invasion(HR=0.190,5% CI:0.020-1.826,P=0.008), Ki67≥14%(HR=0.182,5% CI:0.041-0.801,P=0.024), TNM staging Ⅲ(HR=0.501,5% CI:0.044-5.741,P=0.001) were the independent prognostic factors for TNBC. Conclusion The expression of miR-182 is up-regulated in TNBC, which is correlated with positive regional lymph nodes metastasis and advanced TNM stage. The high expression of miR-182 is one of the independent prognostic factors of TNBC, associated with poor prognosis.
