非受体酪氨酸蛋白激酶Lck与IL-17在急性肺损伤大鼠模型中的表达及意义

摘要:
目的 研究非受体酪氨酸蛋白激酶Lck与IL-17在急性肺损伤(ALI)炎症反应中的表达及意义。方法 气管内滴入盐酸法复制大鼠ALI模型,用免疫组织化学方法(IHC)检测肺组织中Lck和IL-17蛋白表达水平;Western印迹法检测肺组织匀浆中Lck蛋白的表达;用酶联免疫吸附法(ELISA)检测肺组织匀浆和肺泡灌洗液(BALF)中IL-17的含量。结果 正常大鼠肺组织中Lck与IL-17低水平表达,ALI模型建立后6h,其表达显著增高,肺间质中阳性着色的单核细胞明显增多。与对照组比较,ALI组大鼠肺组织匀浆和肺泡灌洗液(BALF)中IL-17含量均显著升高[(102.04±14.23)pg/mg pro vs. (25.16±9.08)pg/mg pro, P<0.01;(66.46±13.12)pg/mg pro vs. (8.19±2.54)pg/mg pro, P<0.01]。结论 ALI大鼠肺组织、肺泡灌洗液、匀浆中Lck与IL-17的表达均上调,两者参与了ALI的炎症反应过程。

Abstract:
Objective To investigate the expression of non-receptor tyrosine protein kinase Lck and IL-17 in acute lung injury(ALI). Methods ALI was induced by inhalation of hydrochloric acid into trachea in rats. Immunohistochemistry was applied to detect the expression of Lck and IL-17 in lung tissue. The expression of Lck in lung homogenate was measured by Western blot. The concentrations of IL-17 in lung homogenate and bronchoveolar lavage fluid(BALF) were detected by ELISA. ResultsLck and IL-17 were expressed at lower level in lungs of normal rats. The levels of Lck and IL-17 were significantly increased 6h after HCI inhalation. The increase of Lck and IL-17-positive mononuclear cells were also found in lung interstitium. The IL-17 levels in lung tissue homogenate and in BALF were higher in ALI group than those in control group [(102.04±14.23)pg/mg pro vs. (25.16±9.08)pg/mg pro, P<0.0l;(66.46±13.12)pg/mg pro vs.(8.19±2.54)pg/mg pro,P<0.0l]. Conclusion The expression of Lck and IL-17 in lung tissue and BALF is up-regulated in ALI rat, indicating that both may be in the inflammatory response of ALI.
