唑来膦酸对兔胫骨骨折愈合的影响

摘要:
目的观察注射用唑来膦酸对兔胫骨骨折愈合的影响。方法 30只新西兰大白兔制成胫骨中段骨折、钢板螺钉内固定骨折模型,随后随机分为实验组和对照组。实验组在术后注射唑来膦酸0.1 mg/kg,对照组注射同样剂量的生理盐水。在骨折模型建立后的第1、2、3、4、6周,通过X线观察骨折愈合情况,并测定血清碱性磷酸酶(alkaline phosphatase,ALP)及血清抗酒石酸酸性磷酸酶5b(tartrate resistant acid phosphatase-5b,TRAP-5b)水平。在建模后的第2、3、4、6周,测定骨折端骨密度。结果实验组血清ALP水平均高于对照组;第3、4周,实验组外骨痂平均面积较对照组大,但差异均无统计学意义(P〉0.05)。同阶段对比,实验组TRAP-5b水平均较对照组低,差异有统计学意义(P〈0.05)。实验组骨密度多数观察时间高于对照组,第4周时两组差异具有统计学意义(P〈0.05)。结论注射用唑来膦酸对稳定固定的兔胫骨横断骨折的愈合可能具有一定的促进作用。

Abstract:
Objective To evaluate the effects of zoledronic acid on the healingof tibial fracture in rabbits. Methods Tibial fractures were induced in 30 NewZealand white rabbits and immobilized with a plate and six screws. The animals were randomly divided intotreatment group and control group: in treatment group zoledronic acid was administered by intravenous injection(0. 1 mg/ kg) a fter fracture,while animals in control group were injected with same volume of normal saline. The states of fracture healingwere examined and then scored by X-ray at week 1,2,3,4 and 6 after fracture. The levels of serum ALP and tartrate-resistant acid phosphatase 5b( TRAP-5b) were assayed,and the bone mineral density at fracture-healingsite was detected at the same time points. Results There were nosignificance differences in serum ALP levels and the area of external callus between treatment and control groups,although there was a trend of increase at week 3 and 4 after fracture. The mean TRAP-5b level of treatment group was lower than that of control group at all time points( P〈0. 05). The average bone mineral density at fracture-healingsite in treatment group was greater than that in controlgroup at week 4( P〈0. 05). Conclusion Systemic administration of zoledronic acid may be effective for healing of tibial fracture under stable fixation in rabbits.
