目的通过观察缺血性脑损伤小鼠脑组织中Parkin、细胞周期素E(Cyclin E)蛋白在用药前后的表达变化,探讨复方蒲黄汤对缺血性脑病的神经保护机制及Parkin蛋白与缺血性脑病的关系。方法 48只雄性C57小鼠随机分成对照组、模型组、药物组,每组各16只。药物组用复方蒲黄汤干预缺血性脑损伤小鼠模型,等量生理盐水灌注胃模型组和对照组小鼠。分别于损伤后0、6、12、24 h断头取脑,免疫组化检测Parkin、Cyclin E的含量。结果模型组Parkin的表达较高,12 h后达到高峰,12~24 h下降,但总体高于对照组(P〈0.05);药物组Parkin整体表达变化类同模型组,但各亚组表达量均低于模型组(P〈0.05),且24 h处下降趋势明显缓于模型组。模型组与药物组Cyclin E的表达都较高,其中12 h处模型组依旧上升,而药物组开始下降,模型组与对照组差异显著(P〈0.05);模型组与药物组在0、24 h时差异有统计学意义(P〈0.05),6、12 h时差异无统计学意义(P〉0.05)。结论复方蒲黄汤可能通过诱导Parkin过表达及抑制Cyclin E表达,达到对缺血性脑病神经元损伤的早期防护。 |

Effects of Compound Puhuang decoction on expression of parkin and cyclin E in brain tissue of mice with ischemic brain injury |

Abstract: |
Objective To investigate the effects of Compound Puhuang decoction on the expression of parkin and cyclin E in brain tissue of mice with ischemic brain injury and its relation toneural functional recovery following is chemic encephalopathy. Methods Forty eight male SPF C57 mice were randomized intosham operation group( n = 16),model group( n = 16) and druggroup( n = 16). The ischemic brain injury was induced by ligation of common carotid arteries for 20 min in model and druggroups. Compound Puhuangdecoction was administrated by gavage in druggroup,while same volume of normal saline was given by gavage in sham and model groups. The mice were sacrificed at0 h,6 h,12 h,24 h after ischemic brain injury( 4 for each batch). The expression of parkin and cyclin E in brain tissue were detected by immuno histochemistry. Results Compared with sham group,the expression of parkin in model group was significantly increased at all time points( P〈0. 05); the peak expression appeared within 12 h and decreased from 12 h to24 h after ischemia; while the expression in druggroup appeared the same trends,but expression levels were significantly lower than that in model group( P〈0. 05). Compared with sham group,the expression of cyclin E was high both in model group and in druggroup; the expression in druggroup decreased from 12 h,while still increased in model group. There were significant differences in cyclin E expression at 0 h and 24 h between druggroup and model group( P〈0. 05). Conclusion Compound Puhuang decoction can up-regulate the expression of parkin and down-regulate cyclin E,which might be associated with its protection of neurons from ischemic injury. |
