目的探讨neuroplastin65对小鼠神经细胞的作用和影响。方法 25~30 g SPF C57BL/6野生雄性小鼠4只,同体质量的NP65(-/-)雄性小鼠4只,分为两组,多聚甲醛和生理盐水灌注固定取材,以14μm冰冻切片。使用尼式染色,对比观察海马神经元的密度。25~30 g的C57BL/6,野生型雄性小鼠8只,同质量NP65(-/-)雄性小鼠8只,线栓法制备局灶性脑卒中模型,缺血损伤2 h后再灌注,7 d以后分别取野生小鼠和NP65(-/-)小鼠大脑组织,灌注固定取材,以4μm石蜡切片,尼氏染色观察两组小鼠神经元存活情况以及TUNEL染色观察神经元凋亡情况。结果 NP65(-/-)小鼠海马和皮质神经元与野生小鼠神经元数量无明显差别;敲除组小鼠的神经元的存活数量较野生型小鼠多;TUNEL染色结果显示,野生小鼠神经元的凋亡数量较NP65(-/-)小鼠多。结论 neuroplastin65的敲除对小鼠神经元产生了影响,这可能会导致小鼠某些神经功能的改变。 |

Abstract: |
Objective To investigate the effect of neuroplastin65(NP65) on neorological function in mice. Methods Brain specimens were taken from 4 SPF C57BL-6 male mice(25-30 g) and 4 SPF NP65(- /-) mice( 25-30 g),and were perfused with normal saline 4% paraformaldehyde.Fourteen μm frozen sections were made for Nissl's stainingand the hippocampal neuron density was observed. Cerebral ischemia was induced by choke line method in 8 C57BL-6 mice(25-30 g) and 8NP65(- /-) mice(25-30 g). The brain was reperfused by pullingthe choke line out 2 h later,the brain samples were perfused by normal saline 4% paraformaldehyde and cut into4 μm paraffin sections. Nissl's stainingand TUNEL stainingwere performed and the sections were observed. Results There were nosignificant differences in the neuronal density of hippocampus and cortex between wildtype mice and NP65(- /-) mice. The paraffin sections showed that more survivinghippocampal neuron cells in NP65(- /-) mice than in wild-type mice,and more apoptotic neurons in wild-type mice than those in NP65(- /-) mice. Conclusion Knock-out of NP65 may attenuate the damage of neuron cells induced by ischemia / perfusion,which may affect on nerve function in mice. |
