Abstract:
Objective To investigate the inductive effect of polycyclic aromatic hydrocarbons (PAHs) on the expression of cytochrome P450 1A1 in A549 cells. Methods PAHs toxicity was screened by MTT assay in pulmonary epithelial A549 cells, and the mRNA and protein expression of cytochrome P450 isozymes (CYP) 1A was analyzed by Real-Time-PCR and Western blot,respectively. Results The PAHs concentration at 0.2 mol/L was determined by cell viability assay for induction screening. Five PAHs, including fluorene, perylene, acenaphthylene, 2,3-benzanthracene, benzo [a] pyrene were identified as CYP1 A1 inducers both in mRNA and protein expression level (P<0.05). Conclusion In this study five PAHs have been identified as CYP1A1 inducers.
