目的探讨罗格列酮(Rosiglitazone,ROZ)体外抑制结肠癌生长的作用。方法体外培养人结肠癌细胞HT-29,不同浓度ROZ(10、20、40、80μmol/L)干预。MTT法测定细胞增殖,透射电镜、TUNEL法及流式细胞术观察细胞凋亡情况,免疫组织化学法检测各组细胞中Bax及Bcl-2的表达。结果MTT法显示ROZ抑制HT-29细胞生长,且具有时间和浓度依赖性,透射电镜、TUNEL法及流式细胞术可观察到ROZ具有诱导结肠癌细胞凋亡的作用,免疫组化法显示经ROZ处理后的结肠癌细胞Bax蛋白表达增强,Bcl-2蛋白表达减弱。结论ROZ体外具有抑制结肠癌细胞生长的作用,与其诱导细胞凋亡有关,可能通过增强Bax的表达及减弱Bcl-2的表达实现。 |

Objective To investigate the inhibitory effect of Rosiglitazone (ROZ) on colon cancer cells in vitro. Methods Cultured colon cancer HT-29 cells were treated by different concentrations of ROZ ( 10, 20, 40 and 80 μmol/L) in vitro. The inhibitory effect of ROZ on the growth of cells was evaluated by MTT; cell apoptosis was detected with TEM, TUNEL and flow cytometry; the protein expression of Bax and Bcl-2 was detected with immunocytochemistry technique. Results The growth and proliferation of HT-29 cells were inhibited significantly by ROZ in a concentration and time- dependent manner. TEM, TUNEL and flow cytometry revealed that cell apoptosis was induced by ROZ treatment. Immunocytochemistry demonstrated that expression of Bax was increased and Bcl-2 expression was decreased after ROZ treatment. Conclusion ROZ can inhibit colon cancer HT-29 cells in vitro, which may be associated with its induction of cell apoptosis, up-regulation of Bax expression and down-regulation of Bcl-2 expression. |
