目的研究载顺铂转铁蛋白长循环脂质体(transferrin modified cisplatin—loaded long circulating liposome,Tf-LDDP)对肺癌A549小鼠移植瘤的抑制作用。方法采用薄膜分散一超声法制备空白脂质体,硫酸铵梯度法包载顺铂,巯基化TfR(transferrin receptor)共价结合顺铂脂质体制备Tf-LDDP。建立肺癌A549小鼠移植瘤模型,以顺铂(cisplatin—loaded long circulating liposome,CDDP)组为阳性对照,生理盐水(normal saline,NS)为阴性对照组,Tf-LDDP为实验组,经尾静脉注射给药,然后观察实验组对肺癌A549小鼠移植瘤生长的影响。结果各组小鼠移植瘤接种成活率均为80%。Tf-LDDP组移植瘤体积明显小于CDDP组[(309.24±160.90)mm3vs(386.33±129.12)mm3,P<0.01];同时Tf-LDDP组移植瘤体积增殖率显著低于CDDP组[(1.38±0.19)I)vs(2.46±0.25),P<0.01]。结论Tf-LDDP对肺癌A549小鼠移植瘤的生长具有一定的抑制作用。 |

Abstract: |
Objective To investigate the inhibitory effect of transferrin-modified cisplatin delivered by long circulating liposome (Tf-LDDP) on the growth of transplanted human lung cancer A549 in mice. Methods The blank liposomes were prepared by the film ultrasound method and cisplatin was encapsulated by ammonium sulfate gradient method. Tf-LDDP was prepared by connecting thiol transferrin with cisplatin-loaded liposome. Human lung cancer A549 cells were inoculated in NOD/ SCID mice. The tumor-bearing mice were injected through tail vein with cisplatin (CDDP) as the positive control, 0.9% NS as the negative control or Tf-LDDP as the experimental group. The tumor weight, size and inhibitory rate were observed in different groups of tumor-bearing mice. Results The survival rate of mice after transplantation of lung cancer A549 was 80%. The tumors size of Tf-LDDP group was smaller than that of CDDP group U (309.24±160.90)mm3vs(386.33±129.12)mm3, P<0.011 ; The rate of increased tumor volume in Tf-LDDP group was lower than that in CDDP group I (1.38±0.19) vs (2.46±0.25), P<0.01 ]. Conclusion Tf-LDDP can inhibit the growth of transplanted human lung cancer A549 in NOD/SCID mice. |
