活性维生素D3对输尿管结扎小鼠肾组织胶原Ⅳ蛋白表达的影响

摘要:
目的通过观察活性维生素D3[1,25-(OH)2VitD3,活性VitD3]对单侧输尿管结扎(unilateral ureteralobstruction,UUO)的CD-1小鼠肾组织胶原Ⅳ蛋白(collagenⅣ)表达的研究,探讨活性VitD3阻止肾组织纤维化作用的机制。方法 42只CD-1小鼠随机分成假手术组(n=6)、UUO组(n=18)、UUO加活性VitD3治疗组(n=18),治疗组再分为0.5、0.25、1.0μg/kg 3个剂量组。UUO组分别于术后(建模后)第3、7、14天处死,其余各组均在术后第7天处死。Western印迹法检测肾脏中胶原Ⅳ蛋白表达量。结果术后第3、7、14天,UUO组肾组织胶原Ⅳ蛋白表达水平都显著低于假手术组,尤其是术后第7天,几乎检测不到胶原Ⅳ蛋白表达。UUO加活性VitD3治疗组的胶原Ⅳ蛋白表达水平显著高于UUO组(P<0.05)。结论活性VitD3对UUO所致CD-1小鼠的肾间质纤维化模型胶原Ⅳ蛋白的表达有一定的促进作用,提示活性VitD3可能具有帮助维持肾小管基底膜完整性的作用。

Abstract:
Objective To investigate the effect of 1,25-( OH) iVitD〗 on renal fibrosis in unilateral ureteral obstruction (UUO) of mice. Methods Forty-two male CD-I mice were randomly assigned to three groups: Sham-operated group (n =6),UUO group (n = 18) and UUO plus 1,25-(OH) zVitD; (0. 5,0. 25,1. 0 (xg/kg) group (n = 18) . Mice of UUO group were sacrificed 3,7,14 d after operation, other groups were sacrificed 7d after operation. The expression of collagen IV in renal tissues was tested by Western blotting. Results Western blotting demonstrated a significant reduction of collagen V production in UUO group, especially at d7 following UUO; the contents of collagen V in renal tissue of 1,25-( OH) iVitDg group were increased compared to UUO group. Conclusion 1, 25-( OH) aVitD; ameliorates renal interstitial fibrosis in the obstructed kidneys,which is associated with the restoration of collagen V production.
