地昔帕明对3-硝基丙酸所致小鼠运动功能障碍的治疗作用

摘要:
目的探讨抗抑郁药地昔帕明对神经毒剂3-硝基丙酸所致小鼠运动功能损伤的治疗作用。方法选用8周龄雄性昆明小鼠36只,随机分为4组:生理盐水对照组、地昔帕明对照组、3-硝基丙酸模型组、3-硝基丙酸模型+地昔帕明治疗组。小鼠神经损伤模型腹腔注射总剂量为320 mg/kg的3-硝基丙酸复制;地昔帕明治疗采用10 mg/kg,连续12 d。采用转棒实验和爬杆实验检验小鼠的运动功能。结果 3-硝基丙酸能够导致昆明小鼠运动功能障碍,模型小鼠在转棒上停留的时间与生理盐水对照组相比明显缩短,爬杆实验转头时间和总时间明显延长。地昔帕明能够明显延长3-硝基丙酸毒性损伤小鼠在转棒上停留的时间,降低爬杆实验的转头时间和总时间。结论地昔帕明对3-硝基丙酸所致的小鼠运动功能损伤具有一定的治疗作用,为研究抗抑郁药治疗与纹状体神经元损害有关的神经精神疾病提供了动物实验依据。

Abstract:
Objective To investigate the neuroprotective effects of desipramine against 3-litropropionic acid-induced behavioral dysfunctions in mice. Methods Thirty six 8^week-old male Kunming mice were randomly divided into 4 groups, normal control group, desipramine control group (desipramine 10 mg/kg for 12 d) , behavioral dysfuction model group and model + desipramine group. The behavioral dysfuction model was induced by intraperitoneal injection of 3-litropropionic acid 320 mg/kg. The locomotor activity and motor coordination were determined by Rotarod test and pole test. Results Systemic 3-NP (320 mg/ kg totally ) treatment significantly reduced locomotor activity and motor coordination as examined by Rotarod and pole-test compared to the vehicle-treated group. Desipramine (10 mg/kg) treatment for 12 d significantly attenuated reduction in locomotor activity and coordination as compared to model group. Conclusion Desipramine can attenuate the behavioral dysfuction induced by 3-litropropionic acid, which indicate that disipramine may be used as antidepressant medication in patients with striatal damages.
