血管紧张素转化酶抑制肽LAP与依那普利改善血管重构的疗效比较

摘要:
目的 比较血管紧张素转化酶抑制肽LAP与依那普利改善血管重构的效应。方法 选取10周龄雄性自发性高血压大鼠(spontaneously hypertensive rat,SHR)为动物模型,随机分为空白对照组(不给药,空白组)、假性实验组(注射生理盐水)、依那普利组(Ena组)、血管紧张素转化酶抑制肽LAP组(LAP组),每组各10只。每日同一时间大鼠腹腔注射药物,实验周期2个月。实验前及实验期间每周测量血压一次,干预结束后于麻醉状态下取大鼠左侧颈总动脉,行形态学研究。结果 NS组与空白组比较,左侧颈总动脉的血管中膜厚度、中膜厚度/腔径、中膜横截面积、中膜细胞平均核面积差异无统计学意义,Ena组与LAP组左侧颈总动脉的上述各指标较空白组比较明显减少,结果差异有统计学意义;LAP组与Ena组比较差异无统计学意义。干预结束后,NS组与空白组比较,左侧颈总动脉的血管中膜胶原纤维面积百分比差异无统计学意义;LAP组及Ena组两种血管的胶原面积百分比均明显低于空白组,LAP组与Ena组相比,差异无统计学意义。结论 血管紧张素转化酶抑制肽LAP能抑制中膜平滑肌细胞肥大,减少中膜胶原纤维增生,从而改善SHR血管重构,改善效果与依那普利相似。

Abstract:
Objective To investigate the effects of angiotensin-converting enzyme inhibitory peptide LAP and enalapril on the vascular remodeling. Methods Forty 10^week-old male spontaneously hypertensive rats (SHR) were randomly divided into four groups with 10 animals in each group: control group (no drug was given), pseudo -experimental group (injection of normal salice, NS group),Enalapril group (Ena group) , angiotensin-converting enzyme inhibitory peptide LAP group(LAP group) . The drugs or normal saline was injected once a day for 2 months. Blood pressure was measured before experiment and once a week during the experiment. After 2 months the left common carotid artery was taken from rats and image analysis for thickness/lumen diameter, medial crosssectional area, mean nuclear area of smooth muscle was performed. Results The vascular medial thickness, media cells of the left common carotid artery in LAP group and Ena group were significantly lower than those in NS and control groups, the difference between LAP group and Ena group was not significant. The vascular area percentage of collagen fibers of the left common carotid artery in LAP group and Ena group were significantly lower than those in NS and control groups. Conclusion The angiotensin-converting enzyme inhibitory peptide LAP can inhibit smooth muscle cell hypertrophy and reduce the proliferation of collagen fibers, thereby improves vascular remodeling of SHR; the effect is similar to enalapril.
