目的 研究UHRF1基因在食管癌组织及癌旁正常组织中的表达及临床意义。方法 对62例食管癌组织及癌旁正常组织应用半定量反转录聚合酶链反应(RT-PCR)检测其中UHRF1 mRNA表达水平;应用Westemblot法检测62例食管癌组织及对应癌旁正常组织中UHRF1蛋白的表达;同时分析UHRF1的表达与患者临床病理特征之间的关系。结果 食管癌组织中UHRF1 mRNA的表达水平显著高于正常组织(P<0.01);Westemb10t结果显示在70.9%(44/62)的食管癌组织中UHRF1蛋白表达较癌旁正常组织高(P<0.001);统计分析结果发现,UHRF1蛋白及mRNA的表达与食管癌患者的淋巴结转移、浸润深度等均无显著性相关(P>0.05),而与食管癌的分化程度呈负相关(r=-0.176,P<0.05),分化程度越低者UHRF1表达越高,各不同分化组间UHRF1表达差异具有统计学意义(P=0.001),UHRF1高表达患者的五年生存率较差。结论 UHRF1在食管癌组织中的表达量显著升高,而且与食管癌分化程度和患者预后相关,可能成为判断食管癌及预后的重要分子生物标志。 |

Objective To investigate the expression of UHRF1 gene in esophagus cancer and its clinicopathological significance.Methods Semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) was applied to detect the expression of UHRF1 mRNA in 62 specimens of esophagus cancer and corresponding adjacent normal tissue;Western blot was performed to evaluate the expression of UHRF1 protein.The correlations of UHRF1 expression with clinicopathological features of esophagus cancer were analyzed.Results The mRNA level of UHRFl was up-regulated in 67.7% (42/62) specimens of esophageal carcinoma as compared with that in matched normal tissues.The protein level of UHRFl was up-regulated in 70.9%(44/62) carcinoma specimens as compared with corresponding normal tissue.UHRFl expression was positively correlated with the differentiation of esophagus cancer(r= -0.176,P <0.05).The median survival was significantly shorter in UHRFl-positive patients than that in UHRFl-negative patients(P = 0.021).Conclusion The expression level of UHRFl is significantly up-regulated in esophagus cancer.UHRFl may be a valuable marker for assessing the prognosis of esophageal cancer. |
