肝癌中Cyclin D1、p16和p21的表达与天然耐药的关系

摘要:
目的 探讨肝癌组织中CyclinD1、p16和p21的表达与天然耐药的关系。方法 128例术前未经化疗的肝癌组织构建组织芯片,其中30例新鲜标本经体外MTT比色分析法检测化疗药物敏感性,免疫组化检测Cyclin D1、p16和p21的表达。结果 30例标本药敏检测后,Cyclin D1、p16和p21在敏感组与耐药组癌组织之间的表达差异无统计学意义(P>0.05);其余98例肝癌组织中,Cyclin D1在P-gp强阳性表达提示的耐药组肝癌中的表达明显高于对照组(P<0.05),p16和p21在两组癌组织中的表达差异无统计学意义(P>0.05)。结论 Cyclin D1可以作为肝癌天然耐药的预测指标;而p16和p21尚不能预测肝癌的天然耐药性,二者与肝癌耐药的关系有待进一步研究。

Abstract:
Objective To study the relationship between the expressions of Cyclin D1,p16 and p21 and intrinsic drug resistance in liver cancer.Methods Immunohistochemistry was employed to detect the expressions of Cyclin D1,p16 and p21 in 128 liver tumors assembled in tissue microarrays.All these patients had not been treated with chemotherepy before operation.Among these cases,30 fresh liver tumors were examined by MTT assay to evaluate chemosensitivitis.Results After 30 cases of liver tumors examined by MTT assay,the positive expression rates of Cyclin D1, p16 and p21 in tumors between resistant group (16 cases) and sensitive group (14 cases) were found with no significant differences (P>0.05) . In the rest 98 cases, the positive expression rate of Cyclin D1 in tumors between P-gp strong-positive-stained group and nonstrong-positive-stained group was significantly different (P<0.05) , but the expression of p16 and p21 in two groups had no differences (P>0.05) . Conclusion Cyclin D1 is a predictive marker of intrinsic drug resistance of liver cancers. P16 and p21 can not predict intrinsic drug resistance in liver cancers and more studies are required to investigate their relationship.
