目的探讨IL-33在大鼠急性胰腺炎(acute pancreatitis,AP)中的作用。方法将64只SD大鼠随机分成急性胰腺炎组(AP组)及假手术组(SO组),每组分4个时间点(术后3、6、12、24h),每个时间点8只大鼠。采用4%牛磺胆酸钠胰胆管逆行注射制备AP动物模型,SO组仅行开腹后翻动胰腺。各组处理后于相应时间点留取大鼠的胰腺组织及静脉血,观察AP病理变化并检测血中AMY、TNF-α和IL-33的变化。结果 AP组的AMY和TNF-α以及病理评分均比同时间点SO组显著升高(P〈0.01),在前6h内随时间逐渐上升(P〈0.05)。在AP发生6h内,AP组IL-33比SO组显著升高(P〈0.01),呈显著上升趋势(P〈0.05),且与TNF-α及病理评分呈正相关,与TNF-α相关系数为0.82(P〈0.01),与病理评分相关系数为0.75(P〈0.05)。6h以后,IL-33呈下降趋势(P〈0.05),24h时降至与SO组相当。结论 IL-33在AP早期上升,且与TNF-α及病理评分正相关,可能在AP早期起促炎因子的作用。 |

The significance of changing of serum IL-33 in acute pancreatitis in rats |

Abstract: |
Objective To study the function of IL-33 in rats with acute pancreatitis(AP).Methods Sixty-four SD rats were randomly divided into AP group and sham operation(SO) group.AP was induced in rats with retrograde infusion of 4% sodium taurocholate into the the pancreastobiliary duct(1ml/kg).The rats were sacrificed at 3,6,12 and 24 h after operation.The samples of blood serum and pancreas tissue were collected at each time point for detecting AMY,TNF-α and IL-33 levels and distinguishing AP pathological score.Results The levels of AMY,TNF-α and pathological score at all time points in AP group were significantly higher than those in SO group(P0.05),and the elevated levels were increased with time during the first 6 hour.The level of IL-33 in AP group was significantly higher than that in SO group at 3,6,12h time points(P0.01) although the level of IL-33 in AP group was dramatic increased during first 6h and then droped rapidly afterward.More interestingly,the level of IL-33 was significantly positively correlated with TNF-α level and AP pathological score at the first 6h time point(r=0.82,P0.01 with TNF-α)、(r=0.75,P0.05 with pathological score).However the level of IL-33 in AP group had no significant difference as compared with that in SO group at 24h time point.Conclusion Elevated IL-33 is significantly positively correlated with TNF-α level and AP pathological score in the early stage of AP indicating IL-33 may play an important role as a pro-inflammatory factor during the early AP development. |
