目的探讨尿多酸肽(CDA-Ⅱ)对NB4、NB4-R1细胞的凋亡效应。方法应用细胞计数法、流式细胞术、DNA ladder实验、RT—PCR观察CDA=Ⅱ处理前后NB4、N134-R1细胞生长增殖状况、凋亡情况及sutpivin基因表达的变化。结果实验组NB4、NB4-R1细胞生长、增殖均受到不同程度的抑制,细胞倍增时间延长。AnnexinV联合PI法细胞早期凋亡实验检测结果显示,随着作用时间的延长,活细胞数量逐渐减少.加药24h后细胞即明显凋亡,随后凋亡细胞进入死亡阶段,死亡细胞比例增加。DNALadder实验显示,加药96h后DNA断裂片断明显增多。RT-PCR实验证明.随着加药时间的延长,survivin基因在mRNA水平上的表达渐弱。结论CDA-Ⅱ可以通过降低survivin基因表达的途径达到促进NB4、NB4-R1细胞凋亡的目的。 |

Abstract: |
Objective To study the effect on apoptosis of NB4,NB4-RI cells by CDA-Ⅱ Methods Apoptosis of NB4,NB4-R1 cells were observed by means of counting the cells,DNA Ladder,flow cytometry,AnnexinV-PI and the expression changes of corresponding apoptosis gene(surviving gene) by RT-PCR. Results Cell growth-inhibition experiments indicated that the experimental groups of" NB4, NB4-R1 cell growth and proliferation are subject to different degrees of inhibition. AnnexinV-PI experiment indicates that the number of living cells reduced gradually with the extension of the role of time. Cell apoptosis was evident by dosing after 24 h. Then apoptotic ceils entered the death phase; DNA Ladder experiments indicated that DNA breakage fragments increased gradually after dosing for 96h. The expression of survivin gene diminuendoe at the mRNA level. Conclusion CDA-Ⅱ can reduce the expression of survivin gene to achieve the purpose of promoting NB4,NB4-R1 apoptosis. |
