应用基因芯片研究藤梨根对胃癌细胞的作用

摘要:
目的研究藤梨根作用于人胃低分化腺癌BGC-823细胞株24h时基因表达谱的改变。方法应用包含105条在肿瘤转化和发生过程密切相关的人类基因cDNA表达谱芯片,检测藤梨根作用于胃癌细胞后基因表达的调控。结果胃癌细胞在藤梨根作用后24h,21条与胃癌发生和转化相关的基因有差异改变,其中4条基因上调,17条基因下调。结论ERBB2、TRAILR2、p53、77、F、CyclinE基因的表达改变可能与藤梨根杀伤和诱导胃癌细胞调亡有关。

Abstract:
Objective To study the effects of Tengligen on gene expression of poorly differentiated human gastric adenocarcinoma cell line BGC -823 after 24 h treatment. Methods cDNA microarray which contained 105 genes associated with tumor cell differentiation and proliferation was applied for the study of gene regulation control of gastric cancer cell after management with Tenglige.Twenty one genes related to tumorigenesis and tumor differentiation detected changed significantly 24 hours after treatment with Tengligen. Among them, 4 genes were up -regulated and 17 genes were down - regulated. Conclusion Alteration of expressions of ERBB-2 , TRAILR2, p53 , TNF and CyclinE genes might be associated with Tengligen treatment which will lead to cell death and cell apoptosis.
