子宫内膜癌患者BRCA1和GPER的表达及其与预后的关系

摘要:
目的探讨子宫内膜癌(endometrial carcinoma, EC)组织中乳腺癌易感基因1(breast cancer susceptibility gene 1, BRCA1)及G蛋白偶联受体(G protein-coupled estrogen receptor, GPER)表达水平及其与病理特征的相关性,并分析其与患者预后的关系。方法采用免疫组织化学染色法检测EC标本中BRCA1、GPER蛋白的表达。χ2检验分析BRCA1及GPER的表达与患者临床病理参数的相关性。BRCA1、GPER与不同病理种类子宫内膜癌的相关性使用多元Logistic回归分析。COX回归方法分析EC预后的独立风险因子。结果BRCA1的表达与EC患者病理类型、组织分化、肌层浸润相关(P<0.05)。GPER的表达与EC患者临床分期、预后相关(P<0.05)。GPER与BRCA1在EC中表达呈负相关性(P<0.05)。国际妇产科联盟分期、年龄、GPER表达情况是EC预后独立危险因素。结论EC组织中BRCA1、GPER的表达与EC部分临床病理特征相关,二者表达呈负相关性,GPER表达水平与EC患者总体预后有关,二者联合检测有望成为EC诊断和治疗的靶点。

Abstract:
ObjectiveTo investigate the expression of breast cancer susceptibility gene 1(BRCA1) and G protein coupled receptor(GPER) in endometrial carcinoma(EC) and their relation with clinicopathological features and prognosis of patients. MethodsImmunohistochemical staining was used to detect the expression of BRCA1 and GPER proteins in EC samples. The correlation of BRCA1 and GPER expression with clinicopathological parameters was analyzed. Multivariate Logistic regression analysis was used to analyze the relation of BRCA1 and GPER expression with pathological types of endometrial carcinoma. COX regression was used to analyze the independent risk factors for EC prognosis. ResultsThe expression of BRCA1 and GPER in EC tissues was correlated with pathological types, tumor differentiation and infiltration to muscular layers of EC(P<0.05); the expression level of GPER was significantly associated with the clinical stages and prognosis of EC patients. The expression of BRCA1 was negatively correlated with GPER expression in EC. FIGO stage, age and GPER were independent influencing factors for prognosis of EC patients. ConclusionThe expression of BRCA1 and GPER in EC tissues was associated with clinicopathological features of EC, and the expression level of GPER was negatively correlated with the overall prognosis of EC patients. The combined detection of BRCA1 and GPER may be used in diagnosis and prognosis for EC patients.
