子宫内膜癌METTL14与核糖体蛋白基因的表达相关性分析

摘要:
目的探究METTL14与核糖体蛋白在子宫内膜癌中的表达相关性。方法通过TCGA数据库TCGA-UCEC获取子宫内膜癌mRNA表达谱数据,采用DESeq2软件包进行基因差异表达(DEGs)分析。采用Metascape数据库对差异表达基因的基因本体论(gene ontology, GO)和京都基因与基因组织百科全书(Kyoto Encyclopedia of Genes and Genomes, KEGG)进行富集分析。使用数据库STRING构建蛋白质互作网络模型分析蛋白质-蛋白质相互作用,应用Cytoscape软件MCODE和cytoHubba模块分析关键基因。结果相较于METTL14高表达的肿瘤组织,低表达METTL14的肿瘤组织中共发现757个差异表达基因(P<0.05),其中665个下调基因,92个上调基因。这些差异表达基因参与的通路主要涉及核糖体功能和氧化磷酸化等,且METTL14与核糖体蛋白相关基因在表达上具有明确的负相关性。结论在子宫内膜癌中,METTL14与核糖体蛋白的mRNA表达具有明确负相关性,提示内膜癌中两者之间可能存在调控作用。

Abstract:
ObjectiveTo explore the relationship between expression of METTL14 and ribosomal protein-related genes in endometrial cancer. MethodsThe transcriptome data of endometrial cancer were obtained from The Cancer Genome Atlas(TCGA) database. DESeq2 was used for analysis of differentially expressed genes(DEGs). Metascape database was used to analyze the gene ontology(GO) terms and pathway enrichment(Kyoto Encyclopedia of Genes and Genomes, KEGG) of DEGs. STRING was used to construct the protein-protein interaction(PPI) network and Cytoscape was used to perform MCODE and cytoHubba analyses for hub gene investigation. ResultsCompared to the tissues with high METTL14 expression, a total of 757 differentially expressed genes were found in the tissues with low METTL14 expression(P<0.05), among which 665 genes were up-regulated and 92 down-regulated. The enriched pathways were mainly involved in ribosomal function and oxidative phosphorylation. There was a significant negative correlation between the expression of METTL14 and ribosomal protein-related genes in endometrial cancer. ConclusionThe expression of METTL14 is negatively correlated with ribosomal protein-related genes in endometrial cancer, indicating that there may be a regulatory network in endometrial cancer.
