Abstract:
ObjectiveTo investigate the expression of ANXA6 gene in human breast cancer and its correlation with clinicopathological features. MethodsParaffin embedded tissue samples were taken from 55 patients with breast cancer from November 2006 to December 2010. Immunohistochemistry was used to detect the expression of ANXA6 gene in breast cancer tissues and para-cancerous tissues. The expression of ANXA6 in breast cancer tissues and para-cancerous tissues was verified by RT-PCR and Western blotting. The relationship between ANXA6 gene expression and clinicopathological features of patients was analyzed. ResultsThe positive expression of ANXA6 gene was 96.3%(53/55) in cancer tissues and 52.7%(29/55) in para-cancerous tissues, respectively(P<0.05). The expressions of ANXA6 and autophagy-related proteins LC3B, P62 and TFEB in cancer tissues were higher than those in para-cancerous tissues(P<0.05). The expression of ANXA6 gene was correlated with tumor size, clinicopathological stage, and PR expression(P<0.05); while not correlated with lymph node metastasis, 5-year survival rate, age, histological grade, ER and HER-2 expression(P>0.05). The expression of ANXA6 was correlated with LC3B and TFEB(P<0.05), but not with P62(P>0.05). The up-regulation rate of ANXA6 gene expression in cancer tissues was correlated with tumor size, lymph node metastasis, clinicopathological stage, 5-year survival rate, age, histological grade, PR expression, HER-2 expression and TFEB expression(P<0.05), but not with ER expression, LC3B and P62 expression(P>0.05). ConclusionANXA6 gene is highly expressed in breast cancer tissues, which is correlated with tumor size, clinicopathological stage, PR expression, LC3B expression and TFEB expression. The up-regulation rate in tumor tissues is corelated with tumor size, lymph node metastasis, clinicopathological stage, 5-year survival rate, age, histological grade, PR expression, HER-2 expression and TFEB expression. It may have some diagnostic value for breast cancer.
