铁死亡在肿瘤中的研究进展

摘要:
铁死亡作为近年来定义的一种调节性细胞死亡方式,以细胞内游离铁增加和脂质过氧化物累积为主要特点。肿瘤是全球第二大致死因素,保护正常细胞的同时有效清除肿瘤细胞是肿瘤治疗的关键。随着铁死亡研究的深入,肿瘤中的铁死亡调控机制不断完善,越来越多的证据表明触发肿瘤细胞铁死亡有望成为新的肿瘤治疗策略。本文从代谢、表观、基因水平对肿瘤中的铁死亡调控机制进行综述,并介绍了基于铁死亡的肿瘤治疗应用,为铁死亡在肿瘤研究和治疗应用方面提供理论基础。

Abstract:
Ferroptosis, as a regulated cell death defined in recent years, is characterized by the increase of free iron level and the accumulation of lipid peroxides in cells. Protecting normal cells while effectively removing tumor cells is critical for cancer therapy. Recently the studies on ferroptosis and its mechanism in tumor regulation has been made progress. There is growing evidence that triggering ferroptosis in tumor cells is expected to become a new strategy for cancer therapy. This article reviews the current progress on the mechanism of ferroptosis regulation in tumors from the aspects of metabolism, epigenetics and genes; and also introduces the application of ferroptosis-based cancer therapy to provide information for tumor research and clinical application in the future.
