摘要:
多能干细胞,包括胚胎干细胞和诱导多能干细胞,均为未分化的细胞,具有自我更新和多向分化潜能,为细胞分化提供了无限的细胞来源,对基础研究和临床疾病治疗研究具有重要意义。然而,多能干细胞的早期谱系分化和调控机制尚不完全清楚。长链非编码RNA(lncRNA)能通过精确控制基因表达网络决定多能干细胞的分化方向,在多能干细胞的分化中发挥重要作用。总结lncRNA在多能干细胞早期分化中的关键作用和调控机制,将有利于更深入地理解多能干细胞的分化过程,为产生下游专能细胞及临床应用提供新的策略。本文将综述近年来lncRNA在多能干细胞早期谱系分化中的作用与机制,为深入理解多能干细胞的分化和调控机制提供指导。

Abstract:
Pluripotent stem cells(PSCs), including embryonic stem cells and induced PSCs, are undifferentiated cells with the potential for self-renewal and differentiation. They provide an unlimited sources for cell differentiation and are of great implication for laboratory research and clinical practice. However, the early lineage differentiation and regulation mechanisms of PSCs are not fully understood yet. Long non-coding RNA(lncRNA) play an important role in the differentiation direction of PSCs by precisely controlling the gene expression network. Knowledge of the function and regulatory mechanism of lncRNA in the early differentiation of PSCs will help to better understand the developmental process of stem cells. This article reviews the recent research progress on the role and mechanism of lncRNA in the early lineage differentiation of PSCs to provide insight for strategies the production of downstream multipotent cells and their potential clinical applications.
