选择性动脉内低温灌注对大鼠局灶性脑缺血再灌注后血脑屏障通透性及血清S100β蛋白的影响

摘要:
目的研究选择性动脉内低温灌注对大鼠局灶性脑缺血再灌注后血脑屏障通透性及血清S100β蛋白的影响。方法构建大鼠大脑中动脉缺血模型,采用不同温度(4、10、20、37 ℃)的生理盐水在恢复血流同时进行灌注,测定各组体温、神经功能评分、脑梗死体积变化,并通过伊文思蓝(Evans blue, EB)含量测定、透射电镜观察比较各组实验动物血脑屏障的功能及超微结构改变,同时通过血清学检测比较S100β蛋白含量的差异。结果研究显示,恢复血流灌注同时进行低温盐水灌注对实验动物核心体温无明显影响,但4 ℃生理盐水灌注较其他温度灌注可显著降低脑温并缩小脑梗死体积、降低EB含量,维持血脑屏障超微结构的完整性,并明显降低血清S100β蛋白的浓度,但对模型建立后48 h神经功能评分无明显影响。结论选择性动脉内低温灌注对大鼠局灶性脑缺血再灌注后血脑屏障的通透性具有显著的保护作用,且4 ℃低温盐水灌注的脑保护作用优于其他温度灌注。

Abstract:
ObjectiveTo investigate the effect of selective intraarterial hypothermia on the permeability of blood brain barrier and expression of S100β in rats with focal cerebral ischemia/reperfusion injury. MethodsThe middle cerebral artery occlusion model was induced in rats. Selective intraarterial hypothermic saline perfusion (4, 10, 20 and 37 ℃) was performed. The neurological function scores, infarction volume, Evans blue (EB) staining, serum S100β level and ultrastructure of blood-brain barrier were compared among different temperature groups. ResultsThe selective intraarterial hypothermic saline perfusion had no significant effect on core body temperature. Hypothermia in 4 ℃ significantly reduced the brain temperature, infarction volume, EB level, serum S100β level, and maintain the integrity of the ultrastructure of blood-brain barrier. It had no influence on the neurological function scores after 48 h. ConclusionSelective intraarterial hypothermia can protect the permeability of blood brain barrier in rats with focal cerebral ischemia/reperfusion injury. The protective effect of 4 ℃ hypothermic saline perfusion is superior to that of other temperature groups.
