1型糖尿病大鼠外周5-HT水平下降与结肠动力障碍的相关性研究

摘要:
目的 研究外周5-羟色胺(5-HT)在1型糖尿病大鼠进程中的变化,并探究其与结肠动力障碍之间的关系。方法 使用链脲佐菌素(STZ)诱导大鼠1型糖尿病,通过免疫荧光与高效液相色谱质谱联用法(HPLS-MS)分别检测糖尿病动物结肠与外周血中5-HT的含量变化;通过Western印迹法检测糖尿病大鼠结肠黏膜层中色氨酸羟化酶1(TPH1)的表达改变;通过检测洋红运输时间考察结肠动力学的改变,观察外源性5-HT(10mg/kg,腹腔注射)对糖尿病大鼠体质量、血糖以及结肠动力障碍的影响。结果 与非糖尿病组相比,STZ诱导的1型糖尿病模型大鼠结肠黏膜层5-HT阳性表达细胞在模型建立后1周(P=0.0003)、2周(P=0.002)和4周(P<0.0001)均有明显减少;血清中的5-HT浓度在模型建立后3周(P=0.0021)和4周(P=0.0009)时明显降低,但黏膜层TPH1表达水平没有明显改变;外源性给予5-HT不影响糖尿病大鼠体质量下降和血糖升高,但可以缓解糖尿病大鼠的结肠动力障碍(P=0.0050)。结论 1型糖尿病动物外周5-HT水平缺乏可能与糖尿病动物结肠动力障碍有关。

Abstract:
Objective To investigate the association of peripheral 5-hydroxytryptamine(5-HT) with colonic dysmotility in type 1 diabetic rats. Methods The type 1 diabetes was induced by injection of streptozotocin(STZ) in rats. The levels of 5-HT in colon tissue and peripheral blood of diabetic rats were examined by immunofluorescence and HPLS-MS, respectively. The expression of tryptophan hydroxylase 1(TPH1) in the mucosa of diabetic rats was detected by Western blot. The colonic motility was evaluated by measuring the transit of carmine red. The effects of exogenous 5-HT(10mg/kg, i.p.) on body weight, blood glucose and colonic motility were measured in diabetic rats. Results Compared with non-diabetic rats, in STZ-induced type 1 diabetic rats, the 5-HT-immunoreactive cells in mucosa of colon was significantly less at 1 week(P=0.0003), 2 week(P=0.002) and 4 week(P<0.0001)after induction of diabetes. The 5-HT level in serum was significantly decreased at 3 week(P=0.0021) and 4 week(P=0.0009) after establishment of diabetes. However, the expression of TPH1 in mucosa was not significantly changed. The exogenous 5-HT improved colonic dysmotility(P=0.0050), while it did not affect the loss of body weight and increase of blood glucose in type 1 diabetic rats. Conclusion The impairment of peripheral 5-HT is associated with colonic dysmotility in type 1 diabetic rats.
