阿尔茨海默病患者脑网络间交互作用异常的静息态fMRI研究

摘要:
目的 利用静息态功能磁共振(functional magnetic resonance, fMRI)探讨阿尔茨海默病(Alzheimer disease, AD)患者脑网络间交互作用异常。方法 采集27例健康老年志愿者及24例阿尔茨海默病患者大脑静息态BOLD-fMRI数据。采用SPM8和RESTplus软件包进行数据预处理,选取属于默认网络(default network, DN)和背侧注意网络(dorsal attention network, DAN)的脑区,提取每个脑区的平均时间序列。为量化DN与DAN之间交互作用,运用功能连接分析法计算每个被试DN和DAN之间(DN-DAN)的功能连接值,对两组受试者的功能连接值进行独立样本t检验,计算两组受试者DN-DAN平均功能连接值。结果 正常老年人DN-DAN平均功能连接值为-0.133,AD患者DN-DAN平均功能连接值为-0.157。与正常老年组相比,AD患者DN-DAN功能连接负值增加(P=0.007)。结论 AD患者DN与DAN之间交互作用存在异常,可为探寻阿尔茨海默病的发病机制提供新思路。

Abnormal interaction between brain networks in patients with Alzheimer’s disease

Abstract:
Objective To investigate the interaction changes of functional brain networks in patients with Alzheimer’s disease(AD) using resting-state functional magnetic resonance(fMRI) data. Methods Twenty-four AD patients and 27 healthy elderly subjects were included in this study. Resting-state bold-fMRI data were acquired and pre-processed with SPM8 and RESTplus software. The brain areas of default network(DN) and the dorsal attention network(DAN) were selected. To quantify the interaction between DN and DAN, functional connectivity analysis were used to calculate the functional connection value between DN and DAN(DN-DAN)of each subject. The independent-sample t-test was used to analyze the functional connection values between two groups. The average functional connection values of the two groups were calculated. Results The average functional connection value of control group and AD group was -0.133 and -0.157 respectively(P=0.007). Conclusion The abnormal interaction between DN and DAN in AD patients may provide new ideas for exploring the pathogenesis of Alzheimer’s disease.
