过表达趋化因子受体2促进脂肪间充质干细胞对皮肤损伤的修复

摘要:
目的 探讨过表达趋化因子受体2(CCR2)是否能促进脂肪间充质干细胞(ADSCs)皮下移植对小鼠皮肤创伤修复的效率。方法 分离培养ADSCs,取3~5代细胞通过流式细胞技术检测ADSCs相关的细胞表面标志物CD73、CD105、CD44和CD31的表达,并分别置于骨、软骨和脂肪诱导分化培养基中进行诱导分化,检测其多向分化潜能。通过慢病毒感染法构建过表达CCR2的ADSCs。在实验小鼠脊背两侧皮肤上分别制备一个面积为0.8cm×0.8cm的机械创面,随机分成3组,分别皮下注射CCR2-EGFP-ADSCs、EGFP-ADSCs和磷酸盐缓冲液(PBS)到小鼠创面4周。每天记录创面愈合情况,并进行病理切片观察,与对照组对比分析。结果 分离培养的ADSCs高表达CD73、CD105和CD44,不表达CD31,具有骨、软骨、脂肪诱导分化潜能。过表达CCR2的ADSCs构建成功。小鼠皮肤创伤造模及移植细胞后,各组小鼠均存活,创面逐日缩小。CCR2-EGFP-ADSCs组、EGFP-ADSCs组和对照组第5、9天的创面愈合率分别为(68±3.1)%和(91±2.1)%、(63±2.7)%和(87±2.1)%、(58±3.5)%和(80±2.9)%。注射了ADSCs的两组愈合率均显著高于对照组(均P<0.05)。病理组织切片显示3组小鼠第9天创面真皮层较正常皮层均有增厚,且CCR2-EGFP-ADSCs组观察到明显的类毛囊结构,多于EGFP-ADSCs组,对照组未观察到类似结构。结论 过表达CCR2可促进ADSCs对皮肤损伤的修复。

Abstract:
Objective To explore if the overexpression of chemokine receptor type 2(CCR2) in adipose mesenchymal stem cells(ADSCs) can promote cutaneous wound healing. Methods The ADSCs were isolated by mechanical separation and enzymatic digestion. The expression profiles of CD73, CD105, CD44 and CD31 on the surface of ADSCs were examined by flow cytometry. The ADSCs from passage 3-5 were used for osteogenic, adipogenic, chondrogenic differentiation assays. The CCR2-overexpressed ADSCs were generated through lentivirus infection. The mice were anesthetized by 10% chloral hydrate and a 0.8cm×0.8cm full-thickness excisional skin wound was made on both sides of the back. Then mice were randomly divided into 3 groups, CCR2-EGFP-ADSCs, EGFP-ADSCs or PBS were subcutaneously injected into the around areas of wound, respectively. The wound healing rate was calculated and the thickness between dermis and epidermis was measured by hematoxylin eosin staining 9 d after model establishment. Results The isolated cells were positive for ADSCs markers, including CD73, CD105 and CD44, and negative for CD31; and had potentials of adipogenic, osteogenic and chondrogenic differentiations when cultured in specific medium. The CCR2 overexpressed ADSCs were generated. All the mice from 3 groups were survived and the wound size reduced gradually. The wound healing rates at d5 and d9 in CCR2-EGFP-ADSCs, EGFP-ADSCs and control groups were(68±3.1)% and (91±2.1)%,(63±2.7)% and(87±2.1)%,(58±3.5)% and(80±2.9)%, respectively(P<0.05). Compared to normal skin, all the three groups showed thicker dermis-epidermis distance. Neogenetic skin appendages were found more in CCR2-EGFP-ADSCs group than in EGFP-ADSCs group, while there was none in PBS group. Conclusion Overexpression of CCR2 in ADSCs can promote cutaneous wound healing.
