原发性大隐静脉曲张中lncRNAs的差异表达

摘要:
目的 比较大隐静脉(great saphenous vein, GSV)曲张血管与正常血管差异表达的lncRNA对大隐静脉曲张的影响,并预测其可能潜在的功能。方法应用表达谱芯片检测6对配对的曲张静脉(varicose vein, VV)和相邻正常节段大隐静脉(normal vein, NV)组织lncRNA表达差异,以qRT-PCR对32例患者样本进行验证;同时以基因芯片lncRNAs和mRNA差异的分层聚类分析、基因本体及通路分析注释差异表达的mRNA通路。结果在6对VV和NV中,存在557个lncRNAs和980个mRNA表达差异;其中6个通路与代谢相关,均得到了qRT-PCR验证。编码-非编码基因共表达网络提示,这6个lncRNA中有4个可能与11个mRNAs相关,且可能与8个代谢通路有关。结论曲张大隐静脉中存在lncRNA异常表达,为lncRNAs生理功能和静脉曲张病理发生提供了新视角。

Abstract:
Objective To identify aberrantly expressed lncRNAs in great saphenous vein (GSV) varicosity and their potential functions. MethodsSix pairs of samples of varicose vein and normal vein were used for lncRNAs expression microarray and 32 samples were used for quantitative qRT-PCR validation. Hierarchical clustering was performed. Gene ontology analysis and pathway analysis were performed to identify mRNA pathways. ResultsTotal 557 lncRNAs and 980 mRNAs that differed between the varicose veins and normal veins were identified in six pairs of samples. These lncRNAs were sub-grouped and mRNAs expressed at different levels were clustered into several pathways with six focused on metabolic pathways. qRT-PCR validated six lncRNAs were consistent with the results of microarray. A coding-non-coding gene co-expression network revealed that four of these six lncRNAs might be correlated with 11 mRNAs and pathway analysis revealed that they might be correlated with another 8 mRNAs associated with metabolic pathways. ConclusionAberrantly expressed lncRNAs for GSV varicosity have been systematically screened and validated and their functions predicted. These findings provide novel insight into the physiology of lncRNAs and the pathogenesis of varicose veins for further investigation. These aberrantly expressed lncRNAs may serve as new therapeutic targets for varicose veins.
