PM2.5对人皮肤成纤维细胞分泌IL-6、TGF-β1的影响

摘要:
目的 探讨大气颗粒物PM2.5对人皮肤成纤维细胞(human skin fibroblast, HSF)分泌IL-6和TGF-β1的影响及胶原多肽(collagen polypeptide, CP)的干预作用。方法 将HSF分为PM2.5组和CP干预组。PM2.5组用0、25、50、100、200μg/ml的PM2.5与HSF培养48h;CP干预组用100μg/ml PM2.5和0、15、30、60μg/ml CP与HSF共培养48h。采用ELISA方法测定细胞上清液中IL-6、TGF-β1含量。结果 随着PM2.5浓度升高(0、25、50、100、200μg/ml),PM2.5组细胞上清液中的IL-6含量也逐渐升高,TGF-β1含量则逐渐下降,两者都存在一定的剂量-效应关系,各炎性因子浓度差异有统计学意义(P<0.05);在PM2.5浓度为100μg/ml时,随着CP浓度升高(0、15、30、60μg/ml),PM2.5+CP组IL-6浓度逐渐下降,TGF-β1含量逐渐增加,两者都存在一定的剂量-效应关系,各炎性因子浓度差异有统计学意义(P<0.05)。结论 PM2.5能促进皮肤炎症,可能与上调IL-6、下调TGF-β1有关;而CP能通过抑制上述作用,缓解PM2.5对皮肤致炎作用。

Abstract:
Objective To investigate the effect of PM2.5 on secretion of interleukin 6 (IL-6) and transforming growth factor β1 (TGF-β1) by human skin fibroblasts (HSF), and the protective effect of collagen polypeptide (CP) from insults of PM2.5. Methods Human skin fibroblasts were co-cultured with PM2.5 0,5, 50,0, 200μg/ml (PM2.5 groups) or PM2.5100μg/ml plus 0,5, 30,0μg/ml CP (PM2.5+CP groups) for 48h. The secretion of IL-6 and TGF-β1 were measured by ELISA method. Results With increasing concentration of PM2.5, the secretion of IL-6 increased and the secretion of TGF-β1 decreased gradually in a dose-effect manner (P<0.05). Compared to PM2.5 group, with increasing concentration of CP the secretion of IL-6 decreased and TGF-β1 increased gradually in PM2.5+CP groups in a dose-effect manner (P<0.05). Conclusion PM2.5 can induce skin inflammation, which is associated with up-regulation of IL-6 and down-regulation of TGF-β1, and CP can inhibit the effect of PM2.5 and alleviate the skin inflammation.
