X射线对人三阴性乳腺癌细胞E-钙黏蛋白基因表达的影响

摘要:
目的 研究不同剂量X射线照射人三阴性乳腺癌细胞,照射后不同时间点对人三阴性乳腺癌细胞E-钙黏蛋白(E-cadherin)基因表达的影响。方法 用X射线分2.5、5和10Gy 3个吸收剂量组照射体外培养的人三阴性乳腺癌HCC1937细胞和MDA-MB-231细胞,采用荧光实时定量PCR技术检测HCC1937细胞和MDA-MB-231细胞经照射后6、12、24和48h的E-cadherin mRNA表达水平,以未照射组(0h组)为对照。结果 HCC1937细胞和MDA-MB-231细胞E-cadherin mRNA的表达在2.5、5、10Gy X射线照射后,随着时间点的后移呈升高趋势。与未照射组相比,除了HCC1937细胞10Gy照射后6h、2.5Gy和5Gy照射后24h及MDA-MB-231细胞5Gy照射后12h外,各时间点差异均有统计学意义(P<0.05)。HCC1937细胞在2.5Gy和10Gy照射后48h,E-cadherin mRNA的表达呈下降趋势,与未照射组相比,差异有统计学意义(P<0.05)。结论 较大剂量的X射线整体可上调HCC1937细胞和MDA-MB-231细胞E-cadherin mRNA的表达,可能诱导了肿瘤细胞凋亡;晚期HCC1937细胞E-cadherin mRNA的表达下降,可能与辐射所致HCC1937细胞的放射生物学效应有关。

Abstract:
Objective To investigate the effects of different doses of X-ray irradiation on the expression of E-cadherin gene in triple negative breast cancer cells. Methods Human triple negative breast cancer MDA-MB-231 and HCC1937 cells were exposed to X-ray Irradiation at a dose of 2.5, 5 and 10Gy for 6, 12, 24 and 48h, respectively. E-cadherin mRNA expression levels in HCC1937 and MDA-MB-231 cells after irradiation were detected with fluorescence real-time PCR technology. Results E-cadherin mRNA expression levels in HCC1937 and MDA-MB-231 cells after irradiation were raised with exposure time. Except HCC1937 cell 6h after 10Gy, 2.5Gy irradiation and 24h after 5Gy irradiation, MDA- MB-231 cells 12h after 5Gy irradiation, there were significant differences at all time points between irradiation and non-irradiation groups(P<0.05). E-cadherin mRNA expression of HCC1937 cells was decreased 48h after 2.5Gy and 10Gy irradiation, compared with those of the non- irradiation group(P< 0.05). Conclusion High dose of X-rays irradiation may upgrade the expression of E-cadherin mRNA in human HCC1937 and MDA-MB-231cells, resulting in cell apoptosis. The decreased expression of E-cadherin mRNA in HCC1937 cells after long exposure to X-ray irradiation might be associated with the biological effect of radiation.
