目的 探讨在可切除的非小细胞肺癌(non-small cell lung cancer, NSCLC)患者血清中循环miRNA的表达。方法 应用实时定量荧光PCR(qRT-PCR)在训练集中检测115名患者和69名健康对照组的术前血清样本中的miR-15b-5p的表达量。用荧光量子点液相微珠在验证集中进一步确认在91名非小细胞肺癌患者和66名健康人群的血清样本中miRNA的差异性表达。用受试者工作曲线(ROC)分析选择最佳的截断值。用卡方检验来分析评估miR-15b-5p关于非小细胞肺癌的诊断价值。结果 miR-15b-5p在非小细胞肺癌中的表达显著上调(P<0.05)。卡方检验结果显示该方法的灵敏度为83.5%,特异度为68.1%。结论 miR-15b-5p可作为可切除的非小细胞肺癌中非侵入性的诊断生物标志物。 |

Serum microRNA as biomarker for detection of early non-small cell lung cancer |

Abstract: |
Objective To investigate expression of circulating miRNA in patients with resectable non-small cell lung cancer(NSCLC). Methods The expression of miR-15b-5p was detected by using quantitative reverse transcriptase polymerase chain reaction(qRT-PCR) in preoperative serum samples of 115 NSCLC patients and 69 healthy controls. The differential expression of miRNA was validated by using fluorescence quantum dots liquid bead array in 91 cases of NSCLC and 66 cases of healthy subjects. Receiver operating characteristic(ROC) analysis was performed and the cut off value of miRNA for diagnosis of NSCLC was selected. The diagnostic value of the miR-15b-5p for NSCLC was analyzed by chi-square test. Results The expression of circulating miR-15b-5p was significantly unregulated in NSCLC patients(P< 0.05). The cut-off value of miR-15b-5p predicted NSCLC with a specificity of 83.5% and sensitivity of 68.1%. Conclusion miR-15b-5p may potentially serve as noninvasive diagnostic biomarkers for diagnosis of resectable NSCLC. |
