miRNA-21-3p对小鼠肝细胞系BNL CL.2体积和增殖的影响

摘要:
目的观察miRNA-21-3p对小鼠肝细胞系(BNL CL.2)体积和增殖的影响。方法小鼠肝细胞系BNL CL.2转染miRNA-21-3p模拟物和抑制物。DiO染膜法和CCK-8法测定miRNA-21-3p对小鼠肝细胞系体积和增殖的影响;荧光定量PCR法检测miRNA-21-3p以及干预该miRNA后与细胞周期相关的基因的表达情况。结果在小鼠肝细胞系,转染miRNA-21-3p的模拟物可以显著增加miRNA-21-3p的表达,转染miRNA-21-3p抑制剂可以显著降低miRNA-21-3p的表达(P〈0.01)。干预miRNA-21-3p不影响细胞的体积。增加miRNA-21-3p的表达显著促进细胞增殖(P〈0.01),而抑制miRNA-21-3p的表达可以显著降低细胞增殖(P〈0.01)。干预miRNA-21-3p,检测细胞周期相关的基因的表达,发现上调该miRNA可以下调Cyclin EP〈0.05),上调Ki67、p15、p16和p18(P〈0.05),且对于CDKN3、Orc6、MCM4和p57没有影响(P〉0.05)。下调该miRNA对于以上基因均没有影响(P〉0.05)。结论miRNA-21-3p对于小鼠肝细胞系体积没有影响,但是可以调控增殖,且它对于增殖的调控效应是部分通过影响周期相关的基因实现。

Abstract:
Objective To investigate the effects of microRNA-21-3p (miRNA-21-3p) on the cell size and proliferation of mouse hepatic cell line BNL CL. 2. Methods BNL CL. 2 cells were transfected with miRNA-21-3p mimics or inhibitor. Cell size was determined with DiO staining while cell proliferation was determined by CCK-8 assay. The expression level of miRNA-21-3p and cell cycle- related genes were detected by Real-Time PCR. Results In BNL CL. 2 cells, miRNA-21-3p mimics increased the expression of miRNA-21-3p, while miRNA-21-3p inhibitor decreased the expression (P〈0.01 ). Although miRNA-21-3p mimics significant increased proliferation of BNL CL. 2 cells, the cell size was not changed;while miRNA-21-3p inhibitors suppressed cell proliferation, miRNA-21-3p mimics significantly down-regulated Cyclin E expression(P〈0.05 ) and up-regulated Ki67, p15, p16 and p18 ( P〈0.05 ) expression; while it did not affect CDKN3, Orc6, MCM4 and p57 expression (P〉0.05 ). The expression of cell cycle-related genes in BNL CL. 2 cells was not changed after transfected with miRNA-21-3p inhibitors (P〉0.05 ). Conclusion miRNA-21-3p promotes proliferation of mouse hepatic BNL CL. 2 cells without affecting cell size, which is associated with regulating some cell cycle- related genes expression.
