| 目的 观察紫草素抑制人肺腺癌A549细胞的增殖及其机制。方法 用不同农度紫草素处理A549细胞，CCK-8方法检测紫草素对A549细胞生长增殖的抑制作用；细胞周期检测试剂盒测定细胞周期的变化;Western blot 法检测磷酸化Akt蛋白（pAkt)的表达。结果 紫草素能够抑制人肺腺癌A549细胞的增殖;诱导A549细胞凋亡；阻滞细胞G1期向S期的发展。pAkt蛋白表达量在紫草素处理48 h后明显减少。结论 紫草素可以抑制肺腺癌A549细胞的增殖，诱导凋亡，机制可能与紫草素抑制PI3K/Akt信号途径有关。 |

| Abstract: |
| Objective To investigate the effect of shikonin on proliferation of human lung adenocarcinoma A549 cells and related mechanisms. Methods A549 cells were incubated with shikonin at different concentration (0.5，1，2 and 4 (xg/ml) for 0，12，24 and 48 h. Cell proliferation was measured by CCK-8 assay ； cell cycle was examined by flow cytometry using propidium iodide staining ； and the expressions of pAkt was tested by Western blot assay. Results The viabilities of A549 cells were significantly inhibited after shikonin treatment. Shikonin can induce A549 cells apoptosis，and blocked the cells developing from G1 phase to S phase. The expression of pAkt was markedly decreased after shikonin treatment for 48 h. Conclusion Shikonin inhibits the growth of A549 cells significantly, which may be associated with inhibition of PDK/Akt signaling pathway. |
