miR-940慢病毒表达载体的构建以及对鼻咽癌细胞的影响

摘要:
目的 构建microRNA-940(miR-940)的慢病毒表达栽体,制备重组慢病毒感染人鼻咽癌细胞5-8F,检测其对细胞迁移和增殖的影响。方法 构建慢病毒载体,包装后感染人鼻咽癌细胞,通过加药筛选方法获得稳定过表达miR-940的人鼻咽癌细胞株5-8F/miR-940。实时定量PCR(RT-PCR)检测miR-940的表达量。MTT测定miR-940对5-8F体外增殖的影响。划痕试验检测miR-940对5-8F迁移的影响。结果 构建稳定过表达miR-940的慢病毒表达栽体。MTT证明miR-940可以抑制5-8F在体外的增殖。划痕试验说明miR-940可以抑制5-8F的迁移能力。结论 miR-940能够抑制5-8F在体外的增殖以及迁移能力。

Abstract:
Objective To construct recombinant lentivirus vector containing miR-940, and to investigate its effect on human nasopharyngeal carcinoma cells. Methods Human nasopharyngeal carcinoma 5-8F cells were transfected with lentivirus vector containing miR-940. The expression of miR-940 in transfected 5-8F cells was detected by RT-PCR, the cell proliferation and migration were assayed by MTT test and wound healing assay, respectively. Results The lentivirus vector was constructed successfully. The transfected 5-8F cells expressed miR-940 stably. Cell proliferation and migration were suppressed in miR-940 transfected 5-8F cells. Conclusion miR-940 can inhibit proliferation and migration of 5-8F cells.
