EphA2与头颈部鳞状细胞癌侵袭、转移及血管生成的相关性

摘要:
目的探讨EphA2与头颈部鱗状细胞癌(head and neck squamou cell carcinomas, HNSCCs)侵袭、转移和 血管生成之间的关系。方法应用免疫组织化学技术检测EphA2在50例HNSCCs组织和30例癌旁黏膜组织中 的表达情况,结合HNSCCs临床病理特征,分析EphA2与HNSCCs侵袭、转移和胂瘤微血管密度(microvessel density, MVD)之间的关系。结果50例HNSCCs组织中,EphA2阳性表达26例,30例癌旁组织中仅2例EphA2呈微弱表达,不同组织中EphA2的表达差异有统计学意义(P<0.05)。EphA2阳性表达与HNSCCs病理分级、侵袭程度和颈淋巴结转移有关(P<0.05),即原发肿瘤分化程度越差、侵袭程度越深,EphA2阳性表达率就越高;有颈 淋巴结转移者EphA2阳性表达率显著高于无颈淋巴结转移者。EphA2阳性者的MVD值(48.68±8.05)显著高于 EphA2阴性者(34.60±6.55)(P<0.05)。结论EphA2促进HNSCCs的侵袭、转移和血管生成,可作为反映 HNSCCs生物学行为的客观指标。

Abstract:
Objective To investigate the correlation of EphA2 expression with invasion, metastasis and angiogenesis in head and neck squamous cell carcinomas (HNSCCs). Methods The expressions of EphA2 and CD34 were detected by immunohistochemical staining in 50 specimens of human HNSCCs tissue and 30 specimens of pericancerous tissue. The correlation of EphA2 expression with the clinicopathologic features and tissue microvessel density (MVD) was analyzed. Results The positive rate of EphA2 protein in cancerous tissues was 52.0% (26/50),significantly higher than that in pericancerous cancer tissues (6.67%,2/30)(x2= 80.000,P=0.000). Expression of EphA2 was positively correlated with histological grade,tumor stage and lymph node metastasis of HNSCCs (x2=65.670,P=0.000;x2=118.235,P=0.000;x2=150.000,P=0.000). MVD in EphA2 positive HNSCCs (48.68±8.05, n=26) was significantly higher than that in EphA2 negative HNSCCs (34.60±6.55,n=24) (t=41.996,P=0.000). Conclusion EphA2 may be involved in the invasive infiltration, metastasis and angiogenesis of HNSCCs, indicating that EphA2 would be used as a marker of biological behavior of HNSCCs.
