NF-κB和STAT3在早期肾癌中的表达及临床意义

摘要:
目的检测核因子KB(nuclearfactor-κB,NF-κB)和信号转导与转录活化因子3(signaltransducerandactivatoroftranscription3,STAT3)在早期肾癌组织中的表达情况,探讨两者在肾癌发生发展中的意义。方法运用免疫组织化学方法检测62例早期肾癌组织和20例癌旁组织标本中NF-κB和STAT3及其磷酸化产物P-NF-κB和P-STAT3的阳性表达情况,分析其与临床、病理参数的相关性;运用聚合酶链技术检测20对肾癌和癌旁组织中NF-κB和STAT3mRNA表达情况,并行统计分析。结果免疫组化结果显示NF-κB、p-NF-κB和P-STAT3在肾癌组织中的表达率分别为62.9%(39/62)、58.1%(36/62)和50.0%(31/62),明显高于癌旁组织的10%(2/20)、0%(0/20)和15%(3/20),差异有统计学差异(P<0.05)。NF-κB和P-NF-κB的表达与临床分期、肿瘤病理分级有相关性。聚合酶链反应(PCR)结果显示肾癌组织中NF-κB mRNA表达率(55.9%)较癌旁组(32.7%)高,差异有统计学意义(P<0.01);肾癌组织中STAT3mRNA表达率(55.3%)较癌旁组(49.7%)高,但差异无统计学意义(P>0.05)。结论NF-κB和STAT3均有望成为肾癌的诊断标志物和治疗靶点,且前者意义更大。

Abstract:
Objective To investigate the expression and the clinic significance of nuclear factor-κB (NF-κB) and signal transducer and activator of transcription 3 (STAT3) in early stage renal cell carcinoma (RCC). Methods The expression of NF-κB and STAT3 protein was determined by immunohistochemistry in 62 samples of RCC tissues and 20 samples of pericancerous tissue. The mRNA expression of NF-κB and STAT3 was detected by RT-PCR in 20 samples of RCC tissues and pericancerous tissue. The correlation of NF-κB and STAT3 expression with clinical and pathological parameters were analyzed. Statistical analyses were performed with SPSS version 13. 0 software. Results NF-κB, p-NF-κB and p-STAT3 in renal cell carcinoma (62.9%, 58.1% and 50.0%) was significantly higher than those in pericancerous tissue (10%, 0% and 15%) (P<0.05). Theoverexpression of NF-κB and p-NF-κB was closely correlated with clinical stage and histological grade (P<0.05). The positive expression rate of NF-κB mRNA in RCC was significantly higher than that in pericancerous tissue (55.9% vs 32.7%, P=0.0015) ; while there was no significant difference in the expression of STAT3 mRNA between two groups (P>0.05). Conclusion NF-KB and STAT3 may be of value in early diagnoses and treatment of RCC.
