miR-145、miR-375在乳腺浸润性导管癌中的表达及临床意义

摘要:
目的 探讨miR-145、miR-375在乳腺浸润性导管癌患者中的表达情况,并做出相应的临床分析。方法 取经病理证实的乳腺浸润性导管癌患者的乳腺癌组织标本及其正常组织各41例,行荧光定量PCR检测其中miR-145、miR-375的表达量,结合临床资料,用SPSS软件分析其阳性表达对临床指导的意义。结果 乳腺癌患者乳腺癌组织中miR-145及miR-375表达明显低于正常组织(P<0.01),miR-145在乳腺癌中的表达异常与病理分期、淋巴结转移、肿瘤大小相关(P<0.01),miR-145的表达与ER、PR无明显相关性,而miR-375的表达与病理分期、肿瘤大小相关(P<0.01),淋巴结转移、PR相关(P<0.05),与ER无明显相关性。结论 miR-145、miR-375在乳腺癌患者中异常表达,癌组织中的表达低于正常组织,这可能是miR-375肿瘤发生过程中担任了抑癌基因的角色,一定程度上可以衡量肿瘤的恶性程度。

Abstract:
Objective To investigate the expression of microRNA-145 and microRNA-375(miR-145 and miR-375)in invasive ductal breast cancer and its clinical significance.Methods Surgical specimens of breast cancer and normal breast tissue were collected from 41 patients with histopathologically confirmed invasive ductal breast carcinoma.The expression of miR-145 and miR- 375 were examined by RT-PCR,and its correlation with clinical data was analyzed by using SPSS software.Results The expressions of miR-145 and miR-375 in breast cancer specimens were significantly lower than those of normal tissue(P<0.01).The expression of miR-145 was closely correlated with TNM stage,lymph node metastasis and tumor size(P<0.01);and was not correlated with ER,PR status.The expression of miR-375 was correlated with TNM stage,tumor size(P<0.01),lymph node metastasis and PR status(P<0.05),and not correlated with ER status. Conclusion miR-145 and miR-375 are down-regulated in breast cancer,which indicates that miR-145 and miR-375 may function as tumor suppressor gene and might be used to evaluate the malignancy of breast cancer.
