兔脂肪间充质干细胞诱导成膀胱移行上皮细胞的研究

摘要:
目的探讨体外诱导兔脂肪间充质干细胞(rabbit adipose tissue-derived stromal cells,rASCs)向膀胱移行上皮细胞分化的可行性。方法实验分混合共培养组、条件培养液组和空白对照组。混合共培养组是将兔脂肪间充质干细胞rASCs(GFP标记)与兔膀胱移行上皮细胞按1:1比例混合培养;条件培养液组是将兔膀胱移行上皮细胞上清液与rASCs一起培养;空白对照组是rASCs与普通培养基常规培养.2周后,用免疫荧光法检测诱导分化的脂肪间充质干细胞中膀胱移行上皮细胞特异性蛋白尿斑素Ib、尿斑素Ⅱ、尿斑素Ⅲ和上皮细胞角蛋白CK18的表达。结果2周后,免疫荧光检测显示,在混合共培养组中,诱导分化的脂肪间充质干细胞表达膀胱移行上皮细胞特异性蛋白尿斑素Ib和上皮细胞角蛋白18、尿斑素Ⅱ、尿斑素Ⅲ没有检测到.而在条件培养液组和空白对照组,上述膀胱移行上皮细胞特异性标记都没有检测到。结论rASCs与兔膀胱移行上皮细胞混合培养可以诱导rASCs向膀胱移行上皮细胞分化。

Abstract:
Objective To investigate the differentiation of rabbit adipose tissue-derived stem cells(rASCs) into bladder transitional epithelial cells in vitro.Methods Bladder transitional epithelial cells and rASCs were isolated and identified from rabbit bladder mucosa and adipose tissue,respectively.The rASCs were labeled with green fluorescence protein.The labeled rASCs were cultured in DMEM medium,or with bladder transitional epithelial cells,or with supernatant of cultured transitional cells,respectively.Two weeks later,expressions of specific bladder transitional epithelial cells differentiation markers were analyzed by immunofluorescence.Results rASCs co-cultured with bladder transitional epithelial cells positively expressed uroplakin Ib and cytokeratin 18,but uroplakin Ⅱ and uroplakin Ⅲ were not detected.Neither of these markers were detected in rASCs cultured with transitional cell supernatant or DMEM medium.Conclusion rASCs co-cultured with bladder transitional epithelial cells can differentiate to urothelial lineage.
