目的 观察冠心病患者外周血单个核细胞Sirts基因的表达。方法 利用RT-PCR从冠心病患者及正常对照组外周血单个核细胞总RNA中扩增出包含Sirts编码区的片段;利用琼脂糖凝胶电泳、实时定量PCR及SPSS分析软件来检测冠心病病例组与对照组单个核细胞Sirts基因表达的区别,以及Sirts基因表达与冠心病危险因子的相关性。结果 冠心病组外周血单个核细胞Sirt1基因的表达(0.75±0.088)明显低于对照组(1.05±0.099),差异有统计学意义(P=0.034)。Sirt1表达与TC、TG、FPG、LDL、ApoA成明显负相关,与HDL成正相关;Sirt2表达与HDL成正相关;Sirt5、Sirt6、Sirt7表达与FPG成负相关(P值均<0.05)。结论 外周血单个核细胞中Sirts基因的低表达与冠心病的发病存在相关性。 |

Abstract: |
Objective To detect Sirts gene expression in peripheral blood mononuclear cells(PBMC) of patients with coronary heart disease.Methods Total RNA was isolated from PBMC of patients with coronary heart disease and healthy subjects(controls),the coding region of Sirtl to Sirtl was amplified by RT-PCR.The expression of Sirts was detected by agarose gel electrophoresis,quantitative real-time PCR.The statistical analysis was performed with SPSS 13.0 software.Results The expression of Sirt1 gene in coronary heart disease patients was significantly lower than that of controls (0.75±0.088 vs 1.05±0.099,P=0.034).Sirt1 gene expression was negatively correlated with TC,TG,FPG,LDL and ApoA levels of patients and positively correlated with HDL levels;while Sirt2 expression was positively correlated with HDL.FPG levels were negatively correlated with Sirt5, Sirt6 and Sirt7 gene expression.Conclusion Coronary heart diseases were likely to associate with low Sirts gene expression in PBMC of patients. |
