类风湿关节炎RF阳性与阴性患者Foxp3表达的差异

摘要:
目的 研究类风湿关节(rheumatoid arthritis,RA)患者类风湿因子(rheumatoid factor,RF)阳性及阴性之间是否存在CD4+CD25+调节性T细胞Foxp3表达的差异。方法RA患者43例.根据RF测定分为RF阳性组和RF阴性组,健康体检者18例为健康对照组。采用流式细胞仪检测RA患者及健康对照组外周血CD4℃D25+T细胞及CD4+CD25+Foxp3+T细胞的表达。结果RF阴性组CD4+CD25+Foxp3+/CD4+T细胞为(2.58±1.68),与对照组(4.23±2.19)相比,差异有统计学意义(P〈0.05);RF阳性组CD4+CD25+Foxp3+/CD4+T细胞为(1.25±1.1),与对照组(4.23±2.19)相比,差异有统计学意义(P〈0.01);RF阴性组与RF阳性组之间CD4+CD25+Foxp3+/CD4+T细胞存在明显差异(P〈0.05)。结论RF阳性与阴性RA患者之间存在调节性T细胞Foxp3表达差异,RF阳性与阴性患者之间存在免疫应答的不同,可能与CD4+CD25+调节性T细胞的功能有关。

Abstract:
Objective To investigate the association of Foxp3 expression with rheumatoid factor (RF) in patients with rheumatoid arthritis (R A). Methods Forty-three cases of R A, including both RF positive and RF negative and 18 healthy subjects were included in this study. Regulatory T cells CD4+CD25+ T cells or CD4+CD25+Foxp3+ T cells in peripheral blood were analyzed by using flow cytometry and comparisons were made between RA patients with RF positive and RF negative and healthy controls. Results The frequencies of regulatory T cells CD4+CD25+Foxp3+ / CD4+ T cells in RF neg ative p ati ents (2. 5 8± 1 . 68) and i n RF po s itive p atients ( 1 . 2 5 土 1 . 1 ) were si gnific antly lower th an th at i n norm al subje c t s ( 4 .2 3±2 . 1 9 ). Furthermore , C D 4+C D 2 5 +Fo xp3+ / C D 4+ T c el l s i n RF n eg ati v epatients were significantly higher than that in RF positive patients. Conclusion RF positive and RF negative RA patients seemed to express Foxp3 differently, which might suggest regulatory activities of T cells could be diffemt in RA patients with or without RF.
