目的建立用酶消化组织块培养兔牙周膜细胞的方法,为下一步的研究做好基础。方法取成年新西兰大白兔牙周膜组织,通过酶消化组织块法原代培养兔牙周膜细胞。取第4代免牙周膜细胞,免疫细胞化学法鉴定细胞组织来源。结果酶消化组织块法成功培养出兔牙周膜细胞,细胞形态呈梭形。免疫细胞化学鉴定波形蛋白染色阳性,角蛋白染色阴性,说明细胞为中胚层来源的牙周膜成纤维细胞。结论酶组织消化块法可以很好地进行兔牙周膜细胞体外培养,且此方法简单易行。 |

Primary culture of rabbit periodontal ligament cells by explants with enzymatic digestion |

Abstract: |
Objective To establish the enzymatic digestion method in cultivating rabbit periodontal ligament cells for further research in future. Methods Primary rabbit periodontal ligament cells were cultured by explants with enzymatic digestion. Immunocytochemical staining was performed for showing its characterization. Results The rabbit periodontal ligament cells were cultured successfully by explants with enzymatic digestion. The ceils were spindie-shaped. These cells were tested positive to antibodies against vimentin and negative to antibodies against cytokeratin by immunocytochemistry analysis indicating themselves to be mesoderm-derived fibroblasts. Conclusion Enzymatic digestion is a simple and practical method for isolation,purification and culture of rabbit periodontal ligament cells in vitro. |
