目的探讨骨肉瘤肿瘤细胞自身血管形成基因表达,为靶向骨肉瘤肿瘤血管治疗提供研究基础。方法利用RT-PCR技术对骨肉瘤肿瘤细胞(MG63、U2OS)血管形成相关基因进行检测,并同内皮细胞(ECV304)相比较。结果骨肉瘤肿瘤细胞和内皮细胞相类似,自身在mRNA水平均能表达VEGF和其受体KDR,受体Tie-2,以及EPHA2,Thrombin,VE-Cadherin等血管形成相关基因。结论骨肉瘤肿瘤细胞表达成血管细胞表型,具有类胚胎表型的逆转,这可能是骨肉瘤富含血供基础,在肿瘤的靶向治疗中,具有进一步的研究前景。 |
