目的 分析成年大鼠小脑基因与新生大鼠小脑基因的差异表达并获取新的表达序列标签。方法 成年大鼠小脑来源的cDNA为受检者(tester),新生大鼠小脑的cDNA为驱动者(driver),进行差减杂交,经克隆、获得成年大鼠小脑差减杂交为。结果 共挑选出54个阳性克隆质粒,测序得到56个不同基因片段序列,其中有16个是大鼠中首次测得的表达序列标签,并被GenBank收录(BG946773~BG946788)。结论 用差减杂交法建立差异表达基因文库可快速筛选未知表达序列标签。 |
